Human Ribosome in situ generated from entry EMD-5224 at the Electron Microscopy Data Bank (EMDB) at PDBe (https://www.ebi.ac.uk/pdbe/emdb/). Subtomogram averaging was used to generate a map with 39Å resolution. A small extra blob of density needs to be removed from the STL before printing.
The model was polished in a sealed small polypropylene tub. Aluminium foil was used to stock the model sticking to the tub. Tissues wetted with a small amount of acetone were placed in the tub (not touching the model) and incubated for 1h at room temperature (21oC). After this time the model was inverted and the process repeated with freshly wetted tissues. Once the additional 1 h incubation was finished the lid of the tub was removed and the model was allowed to dry at room temperature.
The map was downloaded from the EMDB and converted to a surface in Pymol. This was saved as a WRL file. The WRL file was opended in Solidworks and saved as an STL file.
