Summary
I've modified my derivative filament spindle design to accommodate for slightly thicker 1/4" acrylic and made some modifications to reduce the likelihood of "binary failures" including this modification for reducing stress on the t-slots (thanks to tinkerings for pointing this out to me):

Instructions
Same overall construction as the last spindle except that you'll now have to use 20mm or longer bolts to install the pieces that hold the M5 nut in place.
